ABSTRACT

Objective: To establish a inductively coupled plasma mass spectrometry method for the determination of trace cobalt and tungsten in human urine. Methods: The authors used 1% nitric acid solution as diluent in October-December 2021, the sample dilution factor and internal standard element were optimized by single factor rotation experiment, and the difference between the working curve and the standard curve was compared. Results: The method uses working curve to determine cobalt and tungsten in urine, the linear range of this method was 0.0~10.0 μg/L, the correlation coefficient was 0.999 9, the detection limits respectively were 0.005 μg/L (cobalt) and 0.09 μg/L (tungsten), the recoveries of samples respectively were 87.0%~100.2% (cobalt) and 89.4%~104.8% (tungsten), the relative standard deviations respectively were 0.4%~4.4% (cobalt) and 0.6%~3.8% (tungsten) . Conclusion: A simple and rapid method for determination of cobalt and tungsten in urine has been established. This method has the advantages of simple operation, high sensitivity, low detection limit and good stability. It is suitable for determination of cobalt and tungsten in urine of all kinds of people.

ABSTRACT

Objective:To observe the effect of Jinxiangdan (JXD) on NOD-like receptor pyrin domain-containing-3 (NLRP3)/cysteine-dependent aspartate-directed protease-1 (Caspase-1)/interleukin-1<italic>β</italic> (IL-1<italic>β</italic>) signaling pathway in myocardium of rats with myocardial ischemia-reperfusion injury (MIRI) and explore the protective effect and mechanism of JXD against MIRI. Method:Fifty male SD rats were randomly divided into the sham operation group, model group, high- and low-dose JXD groups, and positive drug (Di'ao Xinxuekang) group, with 10 rats in each group. Seven days before modeling, rats in the JXD groups were separately treated with intragastric administration of 0.72 and 0.18 g·kg<sup>-1</sup> JXD tablets, the ones in the sham operation group and model group with the same volume of normal saline, and those in the positive drug group with 1.29 g·kg<sup>-1</sup> Di'ao Xinxuekang. Twelve hours after the last intragastric administration, the anterior descending branch of the left coronary artery was ligated for 30 min and then re-perfused for 60 min for inducing MIRI. ST segment elevation was detected by electrocardiogram(ECG) for model evaluation. The contents of creatine kinase (CK) and lactate dehydrogenase (LDH) in cardiac tissue were measured by colorimetry. Hematoxylin-eosin (HE) staining was conducted for observing myocardial histopathological changes, followed by the detection of cardiomyocyte apoptosis by DNA in situ nick end-labeling (TUNEL) assay. The protein and mRNA expression levels of NLRP3, Caspase-1, and IL-1<italic>β</italic> were detected by Western blot and real-time polymerase chain reaction (Real-time PCR), respectively. Result:Compared with sham operation group, the model group exhibited obviously elevated ST segment (<italic>P</italic><0.01), enhanced CK and LDH activities in the myocardium (<italic>P</italic><0.01), increased apoptotic cardiomyocytes (<italic>P</italic><0.01), and up-regulated NLRP3, Caspase-1, and IL-1<italic>β</italic> protein and mRNA expression (<italic>P</italic><0.01). Compared with model group, JXD at both the high and low doses and Di'ao Xinxuekang significantly lowered the ST segment (<italic>P</italic><0.05,<italic>P</italic><0.01), diminished the CK and LDH activities in myocardial tissue (<italic>P</italic><0.05,<italic>P</italic><0.01), improved the apoptosis of cardiomyocytes (<italic>P</italic><0.05,<italic>P</italic><0.01), and down-regulated the mRNA and protein expression levels of NLRP3, Caspase-1, and IL-1<italic>β</italic> in myocardial tissue (<italic>P</italic><0.05,<italic>P</italic><0.01). The ST segment of ECG in the low-dose JXD group was increased as compared with that in the Di'ao Xinxuekang group (<italic>P</italic><0.05), while the ST segment in the high-dose JXD group was obviously elevated (<italic>P</italic><0.05). Besides, the green fluorescence intensities in the low- and high-dose JXD groups and the Di'ao Xinxuekang group remarkably declined (<italic>P</italic><0.05,<italic>P</italic><0.01). The mRNA and protein expression levels of NLRP3, Caspase-1, and IL-1<italic>β</italic> in the high-dose JXD group were down-regulated (<italic>P</italic><0.05). Conclusion:JXD alleviates MIRI possibly by lowering NLRP3 and IL-1<italic>β</italic> expression and inhibiting cardiomyocyte apoptosis.

Objective To investigate the effect of cervical spinous process fracture with posterior ligamentous complex (PLC) injury on biomechanical stability of the goat cervical spine specimen in vitro,and evaluate the role of posterior structure in maintaining the stability of cervical spine.Methods Twenty-four fresh goat cervical spine C3-6 specimens were randomly and evenly divided into 3 groups:control group (group A),simple cervical spinous process fracture group (group B) and cervical spinous process fracture with PLC injury group (group C).Under loading of 1.5 N · m torque,the range of motion (ROM) in each group was respectively measured under 6 working conditions:flexion,extension,lateral bending and axial rotation,and the ROM differences among 3 groups were compared by using one-way ANOVA analysis.Results Simple cervical spinous process fracture had little effect on the stability of cervical spine and there was no significant difference in ROM between group B and control group (P > 0.05) under all working conditions.Compared with control group,the ROM in flexion,extension and axial rotation significantly increased in group C (P < 0.05),and no significant ROM difference was found in lateral bending between control group and group C (P > 0.05).Conclusions Simple cervical spinous process fracture does not affect the overall stability of cervical spine.Cervical spinous process fracture with PLC injury is more likely to cause cervical instability than simple cervical spinous process fracture,and surgical intervention is required in cervical spinous process fracture with PLC injury.

ABSTRACT

OBJECTIVE: To establish a detection method for chloroprene in human blood by headspace gas chromatography.METHODS: The blood sample was placed directly in the headspace bottle,after the head space automatic sampling,the target object was detected by gas chromatography with electron capture detector,quantified by external standard method.RESULTS: The good linearity range of chloroprene was 0. 00-160. 00 μg / L,and the correlation coefficient was 0. 999 4. The detection limit was 0. 10 μg / L and the minimum detectable concentration in blood samples was 0. 10 μg / L. The with-run relative standard deviation( RSD) was 1. 38%-1. 91%,and the between-run RSD was 2. 11%-3. 08%. The average recovery rate was 89. 00%-93. 88%. CONCLUSION: The method has the features of high sensitivity,wide linear range and simple sample processing,which was suitable for efficient detection for large quantity of samples.

ABSTRACT

OBJECTIVE: To analyze the relationship between the high-temperature working classification and prevalence of hypertension in iron and steel production workers. METHODS: By a convenience sampling method,2 798 workers in an iron and steel production enterprise were selected as study subjects. They were divided into high-temperature working group( 1 070 cases) and non-high-temperature working( control) group( 1 728 cases). The occupational health status was investigated by questionnaire survey and the blood pressure was measured. The wet bulb globe temperature in workplace of high temperature working group was measured,and the high-temperature classification was made based on the time of exposure to high temperature. RESULTS: The prevalence rate of hypertension of 2 798 iron and steel production workers was32. 8%. The prevalence rate of hypertension of high-temperature working group was higher than that of the control group( 41. 3% vs 27. 5%,P < 0. 01). The prevalence rate of hypertension in high temperature working group presented an increasing tendency with the increase of high-temperature working level( P < 0. 01). The multiple logistic regression analysis results showed that excluding the confounding factors of age,body mass index,family history of hypertension,educational background,smoking,alcohol drinking,salt intake and mental stress,it showed that high-temperature working classification was positively correlated with increasing prevalence of hypertension in high-temperature working group( P <0. 01); and the initial exposure age to high-temperature was negatively correlated with the prevalence of hypertension in iron and steel production workers( P < 0. 01). CONCLUSION: High-temperature is an important influence factor to the prevalence of hypertension in iron and steel production workers. Strengthening the management of high-temperature classification is helpful for the prevention and treatment control of hypertension in the iron and steel production workers

OBJECTIVE@#To study the relevance of EGFR gene mutation with pathological features and prognosis in patients with non-small-cell lung carcinoma.@*METHODS@#A total of 297 patients from July 2009 to May 2013 were chosen as objects. EGFR gene mutation were detected with fluorescence quantitative PCR. Relevance of EGFR gene mutation with clinical and pathological features was analyzed, and the prognosis of EGFR- mutant-patients and that of EGFR- wide type-patients was compared.@*RESULTS@#In 297 patients, 136 (45.79%) showed EGFR gene mutation. EGFR gene mutation had no significant relevance with age, gender, smoking history, family history of cancer and clinical stage (P>0.05); there was significant relevance between EGFR gene mutation and blood type, pathologic types, differentiation and diameter of cancer (P<0.05). The difference between prognosis of EGFR- mutant-patients and that of EGFR- wide type-patients was statistical significance (P<0.05).@*CONCLUSIONS@#EGFR gene mutation has significant relevance with pathological features, the prognosis of EGFR-mutant-patients is better than that of EGFR- wide type-patients.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the relationship between epidermal growth factor (EGF) gene polymorphisms at G-61A, R431K, and D784V and susceptibility to silicosis.</p><p><b>METHODS</b>In a case-control study, 116 patients diagnosed with stage I silicosis were included in the case group, and 149 workers without silicosis of the same gender and nationality, exposed to the same nature of dust, and with similar age and cumulative time of dust exposure were included in the control group. Peripheral venous blood was collected, DNA was extracted by salting out, polymerase chain reaction-restriction fragment length polymorphism was used to identify the genotypes at three polymorphic loci of EGF and the allele frequencies, and their distributions in the case group and control group were analyzed.</p><p><b>RESULTS</b>The genotype frequencies of G-61A GG, GA, and AA in the case group were 50.9%, 34.5%, and 14.7%, respectively, and significant differences were found when comparing the data with those in the control group (35.6%,44.3%, and 20.1%), (χ(2) = 6.283, P = 0.048). The distribution frequencies of allele A in the case group and control group were 31.9%and 42.3%, respectively, and the difference between the two groups was statistically significant (χ2 = 5.554, P = 0.018). The risk of silicosis in workers carrying allele G at G-61A was increased by 1.564 times (OR = 1.564, 95%CI: 1.092∼2.024). The genotype frequencies of D784V AA, AT, and TT in the case group were 58.6%, 34.5%, and 6.9%, respectively, versus 65.1%, 31.5%, and 3.4% in the control group, and the differences between the two groups were not statistically significant (χ(2) = 2.278, P = 0.320). The genotype frequencies of R431K GG, GA, and AA in the case group were 56.9%, 39.7%, and 3.4%, respectively, versus 55.0%, 39.6%, and 5.4% in the control group, and the differences between the two groups were not statistically significant (χ(2) = 0.572, P = 0.751).</p><p><b>CONCLUSION</b>The EGF gene polymorphism at G-61A is associated with susceptibility to silicosis, and the risk of silicosis in dust-exposed workers carrying GG genotype is relatively high. No relationship between EGF gene polymorphisms at D784V and R431K and silicosis is found.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the different effects of reinforcing and reducing methods by twirling and rotating the needle on stress-induced hypertension and its mechanism.</p><p><b>METHODS</b>Sixty male Wistar rats were randomly divided into a nomal group (group A), a model group (group B), an acupuncture control group (group C), an acupuncture with reinforcing method group (group D)and an acupuncture with reducing method group (group E), 12 rats in each group. The model of stress-induced hypertension was established by foot-shock and noise stimulation in the other groups except group A. Group C was treated by acupuncture at left "Taichong" (LR 3) with no manipulation, group D with twirling reinforcement method and group E with twirling reducing method at the same acupoint, respectively, and group A and B with no acupuncture treatment. The contents of calcitonin gene-related peptide (CGRP) and nitrogen monoxidum (NO) in blood plasma and hypothalamus after 7 days were observed.</p><p><b>RESULTS</b>After acupuncture treatment, the contents of CGRP in blood plasma and hypothalamus were (375.9 +/- 36.5) pg/mL and (213.6 +/- 50.1) pg/mg in group A, which were higher than (261.0 +/- 20.1) pg/mL and (156.0 +/- 21.8) pg/mg in group B , and (255.3 +/- 33.6) pg/mL and (154.3 +/- 47.3) pg/mg in group C (all P < 0.05), and (465.9 +/- 53.2) pg/mL and (250.74 +/- 47.7) pg/mg in group E higher than those in group C and (283.3 +/- 29.8) pg/mL and (164.6 +/- 47.4) pg/mg in group D (all P < 0.05), and there was no significant difference among group B, C and D (all P > 0.05). After acupuncture treatment, the contents of NO in blood plasma and hypothalamus were (43.7 +/- 5.5) micromol/L and (23.3 +/- 2.9) micromol/g in group B, (46.5 +/- 6.4) micromol/L and (23.1 +/- 3.3) micromol/g in group C, (45.8 +/- 6.7) micromol/L and (25.0 +/- 2.7) micromol/g in group D, which were all lower than (64.5 +/- 9.9) micromol/L and (30.7 +/- 1.6) micromol/g in group A (all P < 0.05), (55.9 +/- 4.6) micromol/L and (30.4 +/- 2.1) micromol/g in group E higher than those in group C and group D (all P < 0.05), and there was no significant difference among group B, C and D either (all P > 0.05).</p><p><b>CONCLUSION</b>Acupuncture with twirling reducing method can increase the contents of CGRP and NO in rats with stress-induced hypertension, thereby causing a fall in blood pressure, however, no significant influence with twirling reinforcing method. Reinforcing or reducing method by twirling and rotating the needle has different efficacy.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the application of temporal fascia flaps in the correction of severe depression deformities at lower eyelids.</p><p><b>METHODS</b>Severe depression deformities at lower eyelid were corrected with temporal fascia flaps pedicled with superficial temporal artery in 9 cases.</p><p><b>RESULTS</b>All flaps survived with good appearance. All patients were followed up for 6-12 months with good long-term results. The donor sites had no obvious scalp scar.</p><p><b>CONCLUSIONS</b>Temporal fascia flap is an optimal choice for correction of the severe depression deformities at lower eyelid. It is easily performed with good result and less donor site morbidity.</p>

ABSTRACT

<p><b>BACKGROUND</b>If the emphysema lesions are not symmetrical, unilateral lung volume reduction surgery (LVRS) can be carried out on the more severe side. The aim of this research was to evaluate the feasibility and effects of LVRS performed simultaneously with resection of pulmonary and esophageal neoplasms.</p><p><b>METHODS</b>Forty-five patients with pulmonary neoplasm and 37 patients with esophageal neoplasm were randomly assigned to group A or group B. In group A, LVRS was performed simultaneously on the same side as thoracotomy. In group B, only tumor resection was performed. The nonfunctional lung area was determined by preoperative chest computed tomography and lung ventilation/perfusion scan. The lung volume removed was about 20% to 30% of the lobes on one side. Preoperative and postoperative indexes including pulmonary function testing variables, arterial blood gas analysis variables, dyspnea scale, 6-minute walk distance, etc., were compared between the groups.</p><p><b>RESULTS</b>There were no surgical deaths in this study. The postoperative forced vital capacity in 1 second, PaO2, PaCO2, dyspnea scale, and 6-minute walk distance were improved significantly in group A, whereas these indexes did not change or decreased slightly in group B.</p><p><b>CONCLUSIONS</b>For tumor patients who have associated emphysema, simultaneous LVRS not only increases the chance of receiving surgical therapy, but also improves the postoperative quality of life of the patient. LVRS has expanded the surgical indication for tumor patients.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the clinical effects of electroacupuncture (EA) at Fenglong (ST 40) on blood lipids.</p><p><b>METHODS</b>Two hundred and four patients of hyperlipidemia were randomly divided into a Fenglong group and a Xuezhikang group, 102 cases in each group. The patients in the Fenglong group were treated with electroacupuncture at Fenglong (ST 40). After arrival of qi, the needles were connected with acupoint nerve stimulator (LH 202 H type, HANS). The primary parameters of EA: for high triglycerides (TG) type, AM 50 Hz, intensity 1 mA, needle-retained time 20 min, twice per week; for high cholesterol (CHO) type, AM 100 Hz, intensity 1 mA, needle-retained time 30 min, thrice per week; for high low-density-lipoprotein (LDL-C) type, the same parameters as the high CHO type except the tolerable and comfortable intensity; for the mixing type, corresponding methods were alternatively used. The patients in the Xuezhikang group received Xuezhikang capsule orally, 2 capsules each time and twice daily, for total 11 weeks.</p><p><b>RESULTS</b>The total effective rates of the Fenglong group and the Xuezhi-kang group were 83.0% and 85.9%, respectively, with no significant difference between the two groups (P > 0.05), and there was no significant differences in the function of regulating blood lipids between the two groups (all P > 0.05). After one month follow-up survey, the total CHO, TG and LDL-C decreased and high-density-lipoprotein (HDL-C) increased, of which there was a significant difference in TG reduction (P < 0.05). There were no relapses in both groups.</p><p><b>CONCLUSION</b>EA at Fenglong (ST 40) can effectively regulate blood lipids with a better after-effect, which can be applied as a safe and effective method to replace medication for regulating blood lipids.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To assess the role of Zhongji (CV 3) in treatment of benign hyperplasia of prostate.</p><p><b>METHODS</b>Multi-central, randomized, controlled, single bland clinical method was adopted, and 276 cases were divided into an electroacupuncture (EA) group and a medication group, 138 cases in each group. The EA group were treated with EA at Zhongji (CV 3) and the medication group with oral administration of Qianliekang tablets. After treatment of 1 course, their therapeutic effects and changes of international prostate symptom (I-PSS) cumulative score, life quality index (L) cumulative score, nocturia times, urine stream state, lower abdominal symptom, maximal volume of urine flow, residual urine volume, prostatic volume, etc. Were assessed in the two groups.</p><p><b>RESULTS</b>The total effective rate was 96.4% in the EA group and 86.2% in the medication group, the former being better than the latter (P<0. 01); the two groups were effective in improvement of international prostate symptom (I-PSS) cumulative score, life quality index (L) cumulative score, nocturia times, urine stream state, hypogastrium symptom, maximal volume of urine flow, residual urine volume, prostatic volume, etc. with the former better than the latter.</p><p><b>CONCLUSION</b>Acupuncture at Zhongji (CV 3) has a significant therapeutic effect for treatment of benign hyperplasia of prostate.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effects of different parameters (frequency, intensity, needle-retained time and treatment interval) of electroacupuncture at Fenglong (ST 40) for adjusting blood lipids, so as to find out the optimization parameter.</p><p><b>METHODS</b>Fifty-four cases meeting the criteria for hyperlipoidemia were randomly divided into 27 groups with orthogonal design L27 (3(13) ). According to the orthogonal design program they were treated with electroacupuncture at Fenglong (ST 40). Ten sessions constituted one course with a one week's interval between two course. The treatment was given for 2 courses.</p><p><b>RESULTS</b>(1) The parameters of EA at Fenglong (ST 40) for regulating blood lipids in primary and secondary orders are: frequency, needle-retained time, interval of treatment, intensity. (2) The parameters of EA at Fenglong (ST 40) for various programs in regulating various blood lipids are: for TG, frequency AM 50 Hz, needle-retained time 20 ain, intensity 1 mA, twice each week; for TC, frequency AM 100 Hz, needle-retained time 30 min, intensity 1 mA, once every other day; for LDL-C, frequency Am 100 Hz, needle-retained time 30 min, intensity tolerable and comfortable, once every other day.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the differentiative capability of adult human bone marrow mesenchymal stem cells (BMSCs) into Schwann-like cells.</p><p><b>METHODS</b>Bone marrows were aspirated from healthy donors and mononuclear cells were separated by Percoll lymphocytes separation liquid (1.073 g/ml) with centrifugation, cells were cultured in DMEM/F12 (1:1) medium containing 10% fetal bovine serum (FBS), 20 ng/ml epidermal growth factor (EGF) and 20 ng/ml basic fibroblast growth factor (bFGF). Cells of passage 1 were identified with immunocytochemistry.</p><p><b>RESULTS</b>Mononuclear cells separated by Percoll's were passaged 10 times by trypsin/ethylenediaminetetraacetic acid (EDTA) digestion in 40 days, and BMSCs increased about 6x10(7) times in this short period. Immunohistochemistry identified that BMSCs were CD34- and CD31-, but they expressed neuron specific enolase; 0.01%-0.02% of total cells expressed nestin, the marker for neural progenitor cells; 40%-50% cells stained heavily by neurofilament 200; and no glial fibrillary acidic protein (GFAP) positive cells were identified; S100 expression was detected among 0.1%-0.2% cells.</p><p><b>CONCLUSIONS</b>Bone marrow contains the stem cells with the ability of differentiating into Schwann-like cells, which may represent an alternative stem cell sources for neural transplantation.</p>

ABSTRACT

Objective To evaluate the feasibility and efficacy of tumor resection combined with lung volume reduction surgery (LVRS) in pulmonary and esophageal patients.Methods 45 patients with lung tumor and 37 patients with esophageal tumor were di- vided into group A(tumor resection plus LVRS) and group B(tumor resection only) randomly.Group A patients underwent tumor re- section combined with LVRS.The target area was determined according to chest computed tomography and ventilation-perfusion scan- ning.The removed pulmonary tissues made up about 20%~30% of the unilateral lung parenchyma.All patients were followed up for 6~12 months.The pulmonary function,blood gas,dyspnea index and 6-min walk distance were compared.Results There were no death during perioperative period.Forced vital capacity in 1 second,PaO_2,PaCO_2,dyspnea index and 6-min walk distance of the group A were all better than preoperative ones,while group B patients had no statistical difference or slight decrease.Conclusion For some tumor patients with emphysema,simultaneous LVRS might provide better opportunity for surgical treatment,but also im- proved the post-operative quality of life.It widens the operative indication for tumor patients.
